Octet分子互作應(yīng)用文獻(xiàn):國(guó)內(nèi)發(fā)表匯總

目前國(guó)內(nèi)高校和科研院所等單位的Octet互作儀已有不下幾百臺(tái)。那這些單位的使用情況如何?是否產(chǎn)生了很多科研成果呢?廢話少說(shuō),數(shù)據(jù)說(shuō)話!

陳老濕發(fā)現(xiàn)截至2021年第一季度,國(guó)內(nèi)單位發(fā)表的文章就有100多篇引用了Octet數(shù)據(jù)的(約占全球Octet文章總數(shù)20%)。去除掉一些預(yù)印版文章、綜述以及未能下載的文章,陳老濕嘔心瀝血的統(tǒng)計(jì)了其中80多篇文章的單位以及應(yīng)用(所有文章在文末列出)。

第一季度文章發(fā)表單位共有60多個(gè),基本都是Octet現(xiàn)有客戶。涉及藥物研發(fā)、植物、腫瘤機(jī)理等各個(gè)方向。其中中國(guó)藥科大學(xué)、鄭州大學(xué)、上海生物化學(xué)與細(xì)胞研究所在2021年第一季度發(fā)表了至少3篇文章,還有其他11個(gè)單位發(fā)表了至少2篇文章。考慮到還有其他文章也是在上海生物化學(xué)與細(xì)胞研究所中完成的(比如上海巴斯德所),所以是當(dāng)之無(wú)愧的最多產(chǎn)Octet客戶。

文章應(yīng)用統(tǒng)計(jì)
 
2021Q1 國(guó)內(nèi)文章應(yīng)用分布

可見,文章的主要應(yīng)用在于小分子、蛋白(含抗體)等物質(zhì)的互作,占了近7成。特別是小分子化合物的相關(guān)文章達(dá)到了3成,足可以說(shuō)明Octet在檢測(cè)小分子化合物上的表現(xiàn)有了長(zhǎng)足進(jìn)步。
另外,檢測(cè)物質(zhì)還有細(xì)菌、多糖、脂質(zhì)體和納米顆粒,再次證明Octet在可檢測(cè)樣品種類范圍方面處于同類儀器領(lǐng)先地位。
 
 
為什么Octet的客戶能在短時(shí)間內(nèi)發(fā)表那么多文章?
 
1  良好的技術(shù)支持
比如西湖大學(xué)(Octet RED96e)、寧波大學(xué)(Octet K2)和華西醫(yī)院(Octet K2)都是2020年年中完成裝機(jī)培訓(xùn)的儀器,使用僅半年就發(fā)表了至少2篇文章。這個(gè)成就離不開強(qiáng)大的Octet技術(shù)支持團(tuán)隊(duì)的貢獻(xiàn)。比如華西醫(yī)院,盡管地處成都,Octet的應(yīng)用科學(xué)家在半年里就去了不下5次。此外西湖大學(xué)的設(shè)備基礎(chǔ)培訓(xùn),由于參與學(xué)員眾多,只能分成4組輪番上陣。雖然我們的培訓(xùn)老師講的口干舌燥,但的確獲得了很好的效果,因此在裝機(jī)培訓(xùn)后的短短2個(gè)月內(nèi)又舉辦了一次針對(duì)性的高級(jí)培訓(xùn)。
 
據(jù)統(tǒng)計(jì),僅2020年下半年,各種基礎(chǔ)培訓(xùn)班、再培訓(xùn)、在線培訓(xùn)的次數(shù)達(dá)到三十幾場(chǎng),培養(yǎng)了一批技術(shù)精湛的Octet使用者。(本周第三條推送有Octet培訓(xùn)報(bào)名,大家走過(guò)路過(guò)不要錯(cuò)過(guò)!)

2  儀器操作簡(jiǎn)單易學(xué),無(wú)需繁瑣日常維護(hù)
Octet系統(tǒng)擯棄了繁瑣的微流路設(shè)計(jì),使用獨(dú)有的生物傳感器浸入即讀檢測(cè)方式,待測(cè)樣品亦無(wú)需特殊預(yù)處理。即使在使用中發(fā)生操作失誤,也不會(huì)導(dǎo)致儀器的損傷,只需重新啟動(dòng)一下就可以繼續(xù)放心使用!儀器使用后無(wú)需繁瑣的維護(hù)工作,只要定期用酒精清潔一下探頭即可。

3  可檢測(cè)樣本種類范圍廣泛,定量化的分子互作表征技術(shù)
上文的應(yīng)用分布充分展示了可用于Octet檢測(cè)的樣品種類之寬泛,尤其可以滿足某些交叉學(xué)科的要求。由于ELISA和Pulldown等傳統(tǒng)方法一般都有洗滌步驟,導(dǎo)致一些解離快結(jié)合弱的互作無(wú)法檢測(cè);而且操作較為繁瑣,測(cè)試一對(duì)樣品的結(jié)合反應(yīng)往往要花費(fèi)1-2天時(shí)間,重復(fù)性也往往不甚理想。當(dāng)實(shí)驗(yàn)有效性存在問(wèn)題時(shí),也難于發(fā)現(xiàn)原因所在。Octet系統(tǒng)不僅可以彌補(bǔ)這些缺點(diǎn),還可以增加互作檢測(cè)效率與可信度,滿足用戶高通量檢測(cè)的需求,在短時(shí)間內(nèi)同時(shí)平行檢測(cè)多對(duì)樣品的相互作用分析。
